The Human Macrophage Inflammatory Protein-1α (CCL3) (Hu MIP-1α) ELISA quantitates Hu MIP-1α in human serum, plasma, cell culture supernatants or other body fluids. The assay will exclusively recognize both natural and recombinant Hu MIP-1α. Principle of the method The Human MIP-1α solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Le CCL3 (MIP1a, Macrophage Inflammatory Protein-1 alpha, MIP-1 alpha) a un poids moléculaire d’environ 8 kDa, appartient à la famille des cytokines chimiotactiques, et est impliqué dans les effets chimiotactiques et pro-inflammatoires, et l’homéostase. Le CCL3 est produit par des lymphocytes, des macrophages et des cellules dendritiques, et est impliqué dans la réponse inflammatoire des monocytes sanguins et des macrophages tissulaires. En outre, le CCL3 peut exister comme un hétérodimère naturel avec le MIP-1bêta et a démontré une activité antivirale contre le HSV-1. La protéine CCL3 peut se lier avec une haute affinité aux récepteurs CCR1 et CCR5.
