The Human Immunoglobulin A (IgA) Uncoated ELISA Kit contains pre-matched antibody pairs, plates, and reagents for performing quantitative enzyme linked immunosorbent assays (ELISA) to detect and quantify protein levels of human IgA. Wash Buffer and Stop Solution are needed to complete the ELISA reaction and are sold separately. Principle of the method ELISAs are designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody is coated to the bottom of the wells of a microplate, which is an overnight process. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. A sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

Les anticorps de contrôle d’isotype peuvent agir comme des contrôles négatifs pour aider à différencier le signal de fond non spécifique du signal d’anticorps spécifique, car ils n’ont aucune spécificité pertinente pour un antigène cible. Bien que les contrôles d’isotypes soient les plus couramment utilisés dans la cytométrie en flux, ils sont utiles dans d’autres applications telles que l’immunoprécipitation de la chromatine (ChIP), l’immunohistochimie et les déplacements de gel. Les contrôles d’isotypes doivent correspondre aux espèces d’anticorps primaires et à l’isotype afin que le niveau de coloration spécifique par l’anticorps primaire puisse être déterminé avec précision. En cas d’utilisation d’anticorps primaires marqués directement, le contrôle d’isotype fonctionne mieux s’il est conjugué avec le même marquage que l’anticorps de test.
