The Human Interleukin 2 (Hu IL-2) ELISA quantitates Hu IL-2 in human serum, plasma, cell culture supernatants, amniotic fluid, synovial fluid, urine or other body fluids. The assay will exclusively recognize both natural and recombinant Hu IL-2. Principle of the method The Human IL-2 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. L’interleukine 2 (IL-2) est une cytokine immunomodulatrice importante pour la prolifération des cellules T activées, la différenciation des cellules B, les cellules tueuses naturelles, les monocytes et les macrophages. IL-2 signale par le récepteur IL-2 (IL-2R), un complexe protéique hétérotrimérique dont la chaîne gamma est également partagée par l’interleukine 4 (IL-4) et l’interleukine 7 (IL-7). L’expression du gène IL-2 dans les thymocytes matures est monoallélique, ce qui représente un mode de régulation inhabituel pour contrôler l’expression précise d’un seul gène. La perturbation ciblée d’un gène similaire à l’IL-2 chez la souris conduit à une maladie de type colite ulcéreuse qui suggère un rôle essentiel de ce gène dans la réponse immunitaire aux stimuli antigéniques.
